Amplification and detection of <\/strong>Mycobacterium tuberculosis <\/strong><\/em>(MTB)<\/strong><\/span><\/p>\n\n

Wear gloves, laboratory coats, mask.<\/li>\n\t
Clean working surfaces with 1% Sodium hypochlorite, followed by 70% alcohol.<\/li>\n\t

4. Switch on the Truelab device by pressing the red button for two seconds.<\/p>\n\n
5. Select test bay.<\/p>\n\n
6. Select test profile: \u201cMTB\u201d, enter Details (Referred By, Patient ID, Gender, Patient Name, Age).<\/p>\n\n
7. Select the sample type.<\/p>\n\n
8. Press \u201cStart Reaction\". For Truelab Uno Dx, Press the eject button to open the chip tray. For Truelab Duo\/Quattro, the chip tray opens automatically on tapping the \u201cStart Reaction\u201d .<\/p>\n\n
9. Take out \u201cTruenat MTB chip\u201d from the chip sleeve (check desiccant colour. If blue -chip can be used; discard chip if desiccant is pink\/ white which indicates moisture is absorbed).<\/p>\n\n
10. Place the chip on the tray by aligning the registration holes with tray pins without touching the white reaction well. (the white reaction well should face upward).<\/p>\n\n
11. 12. Pipette 6 \u03bcl DNA from the Extraction Chamber (After DNA extraction and purification step) into the microtube.<\/p>\n\n
13. Wait 30-60 seconds for the DNA elute and PCR reagent to mix (clear solution obtained).<\/p>\n\n
14. Use the same tip to pipette 6 \u03bcl treated DNA from the microtube and load on the white reaction well of the chip.<\/p>\n\n
15. Discard the microtube and microtip (1% sodium hypochlorite).<\/p>\n\n
16. Start the test run.<\/p>\n\n
17.The test completes in 35 minutes, press \u2018\u2019RESULT\u2019\u2019 to view the result screen\\<\/p>\n\n

19. If MTB detected, test the same DNA eluate for Rif resistance using MTB-Rif chip. Select the MTB-Rif assay.<\/p>\n\n
20. If the result is Invalid\/ Error, repeat amplification using the same extracted DNA and a new \u201cTruenat MTB chip\u201d. If Invalid again, repeat the test with a fresh sample.<\/p>\n\n
21. Tap \u201cOpen\/ Close Tray\u201d button to eject the chip tray.<\/p>\n\n
22. Lift the chip and discard (1% sodium hypochlorite).<\/p>\n\n
