Brain Heart Infusion (BHI) Agar is a general-purpose medium that is:
- Prepared using dehydrated ingredients
- Suitable to grow bacteria, yeasts and moulds
- Used for reagent quality control and contamination check for liquid culture.
To check contamination in Mycobacteria Growth Indicator Tube (MGIT) broth:
- Visually inspect MGIT tubes for heavy turbidity. Heavy turbidity indicates contamination as Mycobacterium tuberculosis growth appears in form of particles without any significant turbidity.
- Divide the BHI agar plate into four quadrants, sub-culture loopful of turbid growth from MGIT tubes in each quadrant (small streak for each specimen, properly labelled).
- Incubate at 37ºC and observe after 24-48 hours for growth of contaminants.
To check the sterility of reagents/ processed specimens:
- Spread one drop of reagent/ sediment on BHI plate.
- Incubate the inoculated BHI plates at 37℃ for 24-48 hours to check for contaminants.
